Fisetin

1.Fisetin can inhibit cardiac hypertrophy induced by pressure overload, improve cardiac function in vivo and inhibit cardiac hypertrophy induced by phenylephrine (PE) in vitro. In hypertrophic heart and cardiomyocytes, fisetin treatment significantly reduced the level of reactive oxygen species (ROS). In addition, fisetin significantly upregulated the expression of antioxidant genes, including catalase (CAT), superoxide dismutase 1 (SOD1) and heme oxygenase 1 (HO-1). In addition, the CO treatment of N-acetylcysteine (NAC, ROS scavenger) and fisetin had no synergistic inhibitory effect on PE induced cardiomyocyte hypertrophy, indicating that the anti hypertrophy effect of fisetin was mainly related to blocking oxidative stress. Finally, after pressure overload and PE treatment, we found that the signal pathway of hypertrophy, mitogen activated protein kinase (MAPK) and mammalian target of rapamycin (mTOR) kinase were inhibited by fisetin.
